Препараты коллагенов в современной дерматологии и косметологии

Коллаген – основной белок кожи, обеспечивающий ее прочность, упругость и эластичность. С возрастом и под влиянием ультрафиолета синтез коллагена снижается, а деградация усиливается, что ведет к морщинам и потере тонуса кожи. Особенно быстро коллаген теряется у женщин в первые годы менопаузы. Пероральные гидролизаты коллагенов – наиболее изученная форма их использования в косметологии. В желудочно-кишечном тракте они перевариваются до коротких пептидов (например, Pro-Hyp, Gly-Pro-Hyp), которые всасываются в кровь и достигают кожи. Эти пептиды не могут встраиваться в коллагеновые волокна. Они действуют как сигнальные молекулы, стимулирующие фибробласты к выработке собственных коллагенов, гиалуроновой кислоты и других компонентов МКМ. Метаанализы многочисленных клинических испытаний показали, что гидролизаты коллагенов приводят к умеренному, но достоверному улучшению гидратации, эластичности и плотности кожи. Более эффективны в этом отношении инъекционные формы коллагенов, которые к тому же демонстрируют сильное защитное действие против старения кожи. Топические формы коллагенов менее популярны и довольно слабо изучены. Особая ниша их использования – покровные материалы для лечения ран. При любом способе доставки в кожу экзогенные коллагены не могут непосредственно встроиться в коллагеновые волокна дермы, их эффект достигается за счет регуляторного действия на метаболизм фибробластов и иммунных клеток. Особое внимание в обзоре уделено рекомбинантным коллагенам, идентичным человеческим. Их получают в клетках бактерий, дрожжей, растений, насекомых и млекопитающих. Наиболее перспективными биотехнологическими платформами представляются дрожжи (Pichia pastoris), растения (табак, кукуруза) и насекомые (шелкопряд). Рекомбинантные коллагены отличаются высокой чистотой, биосовместимостью, отсутствием риска прионных инфекций, гипоаллергенностью. Одним из новых достижений является использование рекомбинантных коллагенов как идеальных «биочернил» для 3D-биопечати живых моделей кожи. Коллагеновые препараты эволюционировали от простых наполнителей к биологическим регуляторам, стимулирующим собственные регенеративные ресурсы кожи. На практике наибольшего эффекта можно достигнуть, сочетая пероральные гидролизаты природных коллагенов и инъекционные формулы на основе рекомбинантных коллагенов.
